L-Glycine | 1,000 mg | L-glycine is a non-essential amino acid with numerous roles in the human body, especially regarding the nervous system and brain health/cognitive function [1]. Taking L-glycine is important when supplementing with NAC, as endogenous glutathione production requires two biochemical steps—in the first step, cysteine (provided by NAC) is added to glutamic acid to form the intermediate glutamylcysteine. In the second step, glycine is added to glutamylcysteine to form glutathione [2]. N-Acetyl-L-Cysteine (NAC) | 600 mg | N-acetyl-L-cysteine (NAC), often called acetylcysteine, is a modified form of the nonessential amino acid L-cysteine with strong antioxidant and mucolytic ("mucus-ridding") properties. NAC is an orally bioavailable precursor for glutathione synthesis in the body, conferring additional protection against oxidative stress.
The association between oxidative stress, mitochondrial dysfunction, and glutathione deficiency is well-established [3]. As we age and glutathione production declines, the body becomes less resilient to chronic oxidative stress, leading to impaired mitochondrial function, elevated inflammation, and other aging hallmarks [4].
Supplementing GlyNAC helps combat the oxidative stress caused by aging and glutathione deficiency, thereby providing several health benefits [5]. In fact, a recent randomized clinical trial in older adults found that GlyNAC supplementation for 24 weeks significantly increased glutathione concentrations, lowered insulin resistance, enhanced body composition, improved age-associated cognitive decline, bolstered exercise capacity, and promoted endothelial function [6].
Moreover, GlyNAC supplementation is gaining traction for supporting brain health. Preliminary findings in aged mice and aging humans show a significant attenuation of oxidative stress and cognitive decline associated with glutathione deficiency after treatment with NAC and glycine [7, 8]. Emothion® S-Acetyl-Glutathione | 200 mg | Emothion® is an acetylated form of glutathione that overcomes the bioavailability challenges of standard glutathione, translating to greater absorption and efficacy. Often considered the "master" intracellular antioxidant, glutathione is a tripeptide comprised of the amino acids glutamate, glycine, and cysteine. This tripeptide is the most abundant intracellular antioxidant in humans. Numerous studies show that glutathione, particularly in its reduced form, mitigates oxidative stress in virtually every body system [9]. However, glutathione production declines as we age, leaving cells with less capacity to fight oxidative stress. Elevated oxidative stress arising from glutathione deficiency harbors multiple aging hallmarks (e.g. cognitive impairment and mitochondrial dysfunction), while restoring glutathione levels does the opposite, supporting cellular health and lessening the risk of age-related conditions [10]. Moreover, recent findings suggest that glutathione is an important regulator of nitric oxide levels in endothelial tissue (the thin layer of cells that lines the inside of your blood vessels and heart) [11]. Given that glutathione improves nitric oxide bioavailability, may prove useful for overcoming the endothelial dysfunction commonly seen in older adults. |
